Interestingly, it does so not only by influencing whether protein-building instructions encoded in DNA are read, but also by altering proteins that are involved in building other proteins. This fine-tuning may enable the cell to react faster to changing needs or situations.

When they disrupted M. pneumoniae's ability to tweak proteins, Gavin, Bork and colleagues also discovered that disaster doesn't necessarily ensue. As in our own cells, proteins in this bacterium rarely work alone. They interact with each other, work together, or perform different steps in chain reactions. The scientists found that these protein networks have a certain buffering ability: disrupting one protein can affect its immediate partners, but the problems may not propagate throughout the whole network. The scientists hope that mapping the different networks may one day enable them to predict where a targeted disruption might do the most damage, which could eventually provide valuable information for drug design.

If Brewers slugger is right, then drug testing must be overhauled to protect players

PHOENIX - Ryan Braun wants everybody to know that he didn't do it. And that he resents the accusations, speculation and erroneous reports that he did.

No, there is no personal medical issue, he's never had an STD, and he passed 25 other drug tests in his career, including three others in 2011.

He isn't happy with the initial MLB reaction that he beat the 50-game suspension on PED charges on a sample-collection protocol technicality.

And, he's considering all legal options in light of everything that has made the last four months, 'the biggest challenge I've ever faced in my life.''

This wasn't Andy Pettitte standing before gathered media, admitting he had used human growth hormone, saying he was sorry, and asking for forgiveness.

This was Braun vowing during a 25-minute press conference on the field at Maryvale Baseball Park on Friday, "If I had done this intentionally or unintentionally, I'd be the first one to step up and say I did it. I truly believe in my heart, and I'd bet my life that this substance never entered my body at any point.''

So a whole new set of questions needs to be examined.

What we have here is a complete reversal in MLB's never-ending PEDs mess. After the game ignored ever-increasing usage from the late 1980s to early-2000s, we've reached the point where players are tied to a testing process in which ? in Braun's words -- they are, "100 percent guilty until proven innocent ? opposite of the American judicial situation."

Not only was Braun's situation wrongly and unfairly leaked to the media by a still-unknown source, even when the hearing produced a not-guilty verdict, the accusers don't want to accept that verdict.

"The system as it was applied to me in this case was fatally flawed," Braun said.

If, as Braun said, "I tried to handle the entire situation with honor, with integrity, with class, with dignity, and with professionalism, because that's who I am, and that's how I've always lived my life," what exactly went on here?

Braun says he knows full well the feeling of being wrongly accused, but nonetheless intimated a possible scenario.

What we do know is that the sample was taken after NLDS Game 1 on Oct. 1 in Milwaukee, along with those of two other teammates. The next 44 hours ? the length of time from when Braun's sample was taken until it was sent by the collector to the lab ? remains in question in the minds of Braun and his handlers.

Why the delay, when Braun says FedEx offices were open until 9:30 p.m. Saturday ? plenty of time after the game that afternoon? And what happened to the sample in that time ? was it stored properly, was it altered in some way at that point? How did he test three times over the allowable limit after passing all those other tests ? including one taken after he received word on Oct. 19 of the positive test result?

"There are a lot of things that we learned about the collector, and the collection process that made us very concerned and suspicious about what could have happened," Braun said.

We're likely to find out the answers to all these questions soon enough, of course. But regardless if the sample was tampered with maliciously, or just by the result of human error, Braun's reputation severely has been damaged.

Can he restore it? And don't changes have to be made to the collection process? This situation is far from over, and very well could be a precedent-setter.

"We won because the truth was on my side," Braun said. "I was a victim of a process that completely broke down and failed. Today is about making sure this never happens to anyone else who plays this game."

MLB players better hope so, because another ugly can of worms in the ongoing PEDs saga has been opened ? if Braun's words indeed are true.

There seems to be distinct stages of fitness motivation that women go through.

Regardless of how long you?ve been dedicated to your fitness routine, there may still be times when you get bored. Your motivation can be challenged by the familiarity of the routine; after all familiarity breeds contempt. When it comes to health, fitness, and working out, these are highly personal. Our challenges to maintain motivation and keep moving forward will also be personal. This means that we need to find our own way of dealing with this issue. It only makes sense that a personal solution is needed for a personal problem. So you need to find your own sources of inspiration and motivation that mean the most to you, personally. It can be health-related or concerning your physical appearance, or both ? doesn?t matter.

As soon as you catch yourself practicing a harmful habit, do everything you can to reverse it. The key is to be honest with yourself, and what?s going on with you, and then do whatever is necessary to stop this from becoming a real problem. If you just ignore it, then it?s likely that you?ll have a hard time staying motivated in the long run. Once your excitement and desire to get healthy and looking great begins to fade, then it is like starting all over again. Simply decide that you won?t allow this to happen.

As you continue with your work outs, you will eventually be confronted with fitness motivation issues that are peculiar to women. You can help yourself by doing some research and learning what you can about this issue. There is a lot you can do to retain overall control on the situation and effectively deal with it.

You can even take a proactive approach and have measures in place that can serve to prevent motivation issues from occurring in the first place.

There's a scene in Pixar's Finding Nemo when Dory, a yellow-finned regal tang, injures herself in a tug-of-war over a snorkel mask. A tiny plume of blood curls away from Dory's face into the water around her, where it is sucked into the nostrils of Bruce, a "vegetarian" shark who immediately recants his no-sushi policy. (Fortunately, Dory escapes.) Scientists have known for some time that many ocean predators relish the scent of an injured fish, whereas fish that are more likely to end up as a meal flee from the same scent. Now, researchers think they have pinpointed the key chemical in fish skin that warns nearby fish of danger?a chemical related to a supplement some people take for joint pain.

In the 1930s Austrian animal behavior scientist Karl von Frisch accidentally injured a minnow in a tank. He noticed that the other fish in the tank began alternately darting back and forth and freezing in place?classic predator-evading behavior. Subsequent experiments established that the frightened fish were responding to chemicals released from the skin of their injured peer?a cocktail dubbed schreckstoff, which is German for "scary stuff."

For decades, the chemistry of schreckstoff remained unknown. In the 1970s and '80s some scientists discovered that exposing fish to a chemical known as hypoxanthine-3-N-oxide (H3NO) frightened them in the same way as schreckstoff, albeit to a lesser degree. H3NO, they concluded, was probably the active compound in schreckstoff. But there was a problem with that idea: scientists had never reliably detected H3NO in fish skin. Instead, some researchers proposed, H3NO may mimic the genuine active compound.

Now, Ajay Mathuru of Neuroscience Research Partnership in Singapore, Rainer Friedrich of the Friedrich Miescher Institute for Biomedical Research in Switzerland and their colleagues think they have isolated the key ingredient in schreckstoff?a sugarlike molecule named glycosaminoglycan (GAG) chondroitin. Their findings appear online in the February 23 issue of Current Biology.

Friedrich and his colleagues studied schreckstoff extracted from ground-up zebra fish, which are closely related to minnows and catfishes. The researchers performed a series of tests on the schreckstoff to identify the active compound. For example, destroying all the proteins in the solution did not diminish schreckstoff's ability to frighten fish, suggesting that the active compound was not a protein. In another test, the researchers dipped a column of wheat germ agglutinin?a sugar-binding protein that protects wheat from insects and pathogens?into schreckstoff and introduced fish to the compounds the wheat germ had soaked up. The fish behaved as though they were escaping a predator: they stayed near the bottom of the tank and rushed from one spot to another or they swam very slowly for a short time and then shot off in another direction over and over again. Perhaps the active compound in schreckstoff was similar to the sugars sponged up by the wheat germ column.

As expected, Friedrich and his colleagues detected glycosaminoglycans in zebra fish schreckstoff, in particular glycosaminoglycan chondroitin. Glycosaminoglycans are long, sugar-like molecules that exist in various forms in living creatures, most notably in cartilage and other connective tissue. When the researchers broke down the glycosaminoglycan chondroitin in schreckstoff with enzymes or rendered chondroitin inert with antibodies, they robbed schreckstoff of its power to frighten. Further, exposing fish to chondroitin alone triggered darting, slow swimming and bottom-dwelling?but not as effectively as undiluted schreckstoff.

"It's really nice to have biochemists seriously looking at the chemistry of chemical alarm cues in fish," says Brian Wisenden of Minnesota State University Moorhead, who has studied schreckstoff in minnows but was not involved in the new study. "We've basically been working with a mystery substance."

Schreckstoff also appears to activate a specific region of the olfactory bulb, the part of the brain that processes odors. Anatomically unusual neurons called crypt cells are woven into this part of the olfactory bulb?neurons whose function remains a mystery. Friedrich and his colleagues suggest that these cells are specifically dedicated to sensing schreckstoff. When they exposed fish to other odors, such as bile acids and amino acids, the neurons in this part of the brain did not respond?they were only interested in schreckstoff.

Ever since the discovery of the scary stuff, scientists have wondered why fish would evolve to release a chemical alarm when injured. Sure, it is a nice gesture toward other fish, but how does the injured animal benefit? Warning siblings and cousins of danger helps protect one's own genes?a strategy known as kin selection?but schools of fish are not necessarily populated by members of the same family.

Another explanation is that there is no real benefit to the injured fish. Instead, evolution has favored fish that recognize the chemicals naturally released by injured comrades?just as some plants have evolved to recognize chemicals released by neighbors under attack. In both these situations, injured organisms are not intentionally releasing chemicals. In fact, they would rather keep those useful substances to themselves. The volatile compounds that escape from injured plants are usually toxins stored to defend against herbivores. And some evidence suggests that the GAGs in schreckstoff play an important role in the skin's immune system. As a natural consequence of physics, these molecules are always released into the water when a fish is injured. In this way, certain compounds are consistently associated with injury?and any fish that recognizes that link has a better chance of escaping predators and surviving. Over time, evolution establishes these compounds as reliable signals of danger. Today, all zebra fish?and indeed many fish species?are born with brains wired to learn that a whiff of schreckstoff is their cue to hightail it.

Regulations Harm Small Business and Protects Corporations :

The prospects for conducting commerce are never an easy task. The hurdles to start a business much less stay competitive demands the greatest skill and fortitude. Innovation and inspiration often is the best course for those bold enough to become an employer.

The idea that a level playing field exists for all comers is preposterous. The entire macrocosm for business rests upon separating your enterprise from that of your rivalries. Such is a basic lesson for those brave or foolish enough to enter the arena.

Courtney Rubin?cites the following in Inc. Magazine,

?Businesses with 20 employees or fewer pay 36 percent more than their larger counterparts (defined as those with 500 or more employees), says the?report?? called ?The Impact of Regulatory Costs on Small Firms? ? from the SBA?s Office of Advocacy. This is because a lot of costs are fixed ? the same whether you have two employees or 2,000. Total annual cost of following the rules for a small business: $10,585 per employee, or about $2,830 more than big business. Businesses with 20 to 499 employees paid about $7,454 per employee, or about $300 less than the largest companies.

The report estimates that 89 percent of all firms in the U.S. employ fewer than 20 workers. By comparison, large firms account for only 0.3 percent of all U.S. firms.

Says the report:

?If federal regulations place a differentially large cost on small business, this potentially causes inefficiencies in the structure of American enterprises, and the relocation of production facilities to less regulated countries, and adversely affects the international competitiveness of domestically produced American products and services.?

The screams for jobs, jobs and jobs would give the hint that federal, state and local business policy would favor the productive engine of employment.

However, in the real world of political influence and favoritism only the well connected get the advantages.

Government regulations are meant to stifle competition. The legislative process graces those who are well connected, financially heeled and schooled in the art of writing the regulations. Few small businesses have a legal department or experienced lobbyists.

In Big Business and Big Government,?Timothy P. Carney?writes,

?As the federal government has progressively become larger over the decades, every significant introduction of government regulation, taxation, and spending has been to the benefit of some big business.?

Mr. Carney presents compelling evidence on the history of this axiom, in his article. The net result from this covert partnership of interest and rewards is the never ending campaign contribution cycle that finances every election.

The small businessman seldom has the resources or crony relationships to wage off the grand strategies of the giant corporate model.

Their advantage stems not from mastering sound and creative business practices. On the contrary, the major corporations use their brute force to buy or stamp out any contender that dares compete for market share.

Access to capital or the lack thereof, dooms most small businesses. The burden of regulations only compounds the severity of the survival rate and burns up reserved funds which often cannot be replenished.?How can small business compete? ? offers this insight.

?Small business can?t control mass-market designs or brands, but we are well-placed to do what big business can?t:?Get inside the hearts and minds of our customers.?

As true as that advice resonates, the regulation landmines prevent small businesses from operating on a scale that can challenge all the advantages of the state sponsored conglomerates.

The hard truth is that free enterprise is dead and in its place is an administered economy designed to suppress individually owned and managed businesses.

The prospects of reestablishing a political atmosphere that favors small business as the primary mechanism of job creation is remote as long as the transnational global and corporatist culture exists.

Bigger is not better in most cases. Bigger usually means there are fewer companies in the same industry, accompanied with shrinkage in good paying jobs.

Theodore F. di Stefano?suggests four steps that small businesses need to focus upon,

1. Creating a Niche

A niche is a special quality or group of qualities that sets the small business apart from its larger competitors. It has also been described as a small, specialized business market.

2. Employee Training

If a small business is going to act as though the customer is truly special, its employees must be trained accordingly. Also, they must work for managers whom they respect and who respect them. They must not be put on the ?floor? to meet customers until they are thoroughly familiar with their product, be it food, auto services or any other product.

3. Management Philosophy

The owners of a small business must know the goals (mission) of their business and how they intend to achieve these goals. They should be clear about what segment of the market is their target and how they intend to appeal to that segment.

4. Good In-House Financial Management

You must be particularly aware of your current and projected cash position. And, you should certainly create a realistic annual budget for your company that serves as a financial road map for the future.

Now these common sense suggestions may assist in certain instances, but in a service economy, living wage jobs are rare at best.

The regulations that drive business offshore also destroy a viable income scale. Reinstituting an American industrial and manufacturing base is a necessary step to climb out of this deep hole.

The regulatory climate must reflect policies that will benefit American workers. Open borders, that encourage illegal immigration, are a conscious regulatory policy that displaces domestic employees.

The regulations that slant and foster corporatist preference is the new feudalism. Is it not time to put Americans back to work?

Without a political will to champion free enterprise and replace the corporate-state, only more suffering will thrive. It is up to the public to make this challenge the centerpiece for the 2012 elections.
